Heat shock protein 90 of Bonamia ostreae: characterization and possible correlation with infection of the flat oyster, Ostrea edulis ArchiMer

In this study we described the cytosolic HSP90 of Bonamia ostreae, an intracellular parasite of Ostrea edulis haemocytes. The complete open reading frame was assembled by Rapid Amplification cDNA Ends reactions on cDNA of B. ostreae infected haemocytes. HSP90 amplification was corroborated in infected oysters and B. ostreae purified cells. The functionality of the HSP90, studied by inhibitory assays with radicicol, suggests that this protein may play a role in haemocyte invasion. Our results inform the molecular basis that governs B. ostreae-O. edulis interactions.

Herpes-like virus infecting Japanese oyster (Crassostrea gigas) spat ArchiMer

During the summer of 1993, abnormal sporadic mass mortalities (80-90%) and morbidities occurred among four batches of young Japanese oyster (Crassostrea gigas) spat from the French Atlantic coast. These mortalities were only observed in July. Samples of affected animaIs were examined by electron microscopy. This paper reports the presence and appearance of herpes viruses in these samples.

Infection of clams by Perkinsus olseni as an example of the new French Surveillance Programme ArchiMer

The French network for the surveillance and monitoring of mollusc health (REPAMO) was officially set up in 1992 to control the shellfish status along the French coasts according to the European directives 91/67/EEC and 95/70/EC. The objectives are 1) the surveillance of notifiable diseases present in France (Bonamiosis and Marteiliosis of flat oysters Ostrea edulis), 2) the surveillance of health status of cultured and wild populations of molluscs, 3) the study of abnormal mortality and 4) the control of exchanges from European and third countries. Infection with the protozoan parasite Bonamia ostreae modifies in vitro haemocyte activities of flat oyster Ostrea edulis ArchiMer

Bonamia ostreae is an intracellular protozoan parasite, infecting haemocytes of the European flat oyster Ostrea edulis. Oyster defence mechanisms mainly rely on haemocytes. In the present study in vitro interactions between parasites and flat oyster haemocytes were investigated using flow cytometry and light microscopy. Haemocyte parameters including: non specific esterase activity, reactive oxygen species (ROS) production and phagocytosis were monitored using flow cytometry after 2 hour cell incubation with live and dead B. ostreae. Bonamia ostreae is an intrahemocytic parasite that has been responsible for severe mortalities in the flat oyster Ostrea edulis since the 1970́s. The Pacific oyster Crassostrea gigas is considered to be resistant to the disease and appears to have mechanisms to avoid infection. Progeny of Crassostrea virginica oysters, introduced into France in 1992, were reared during 1992 and 1993, to test their growth performances. These assays were performed to investigate the possible replacement of Crassostrea gigas with a non indigenous species, Crassostrea virginica. During the summer of 1993, sporadic mass mortalities (80-90%) occurred among young, Crassastrea virginica, oysters reared in the IFREMER nursery in La Tremblade (Charente Maritime).

The Ifremer Repamo network (REseau de PAthologie des MOllusques) is the national surveillance network in charge of monitoring marine mollusc health on French coastlines. It is commissioned to enforce the official regulations and to provide a public service, under the supervision of the competent authority (DGAL Veterinary Directorate) in the Ministry.of Agriculture and Fisheries. Its goals are to prevent the introduction and the dissemination of infectious agents, in particular pathogens involved in reportable diseases, and to monitor the changes in the pathogens already present on the French coasts.

Progeny of Crassostrea virginica oysters, introduced into France in 1992, were reared at 3 different locations. A survey was taken of these young C. virginica oysters during 1993. Histological and electron microscopical examination revealed a parasite interpreted as a Martelia-like protozoan in 1 oyster. The structure and the initial stages of the developmental cycle of this parasite are described for the first time in American oysters. However, no mature spores were detected. Thus, exact identification of the protozoan is difficult. This study indicates that American oysters could be infected with Marteilia spp. during cohabitation with Marteilia-infected native bivalves in France.

Molecular characterization of parasites of the genus Perkinsus present in clams from French producing areas ArchiMer

Molecular detection and quantification of the protozoan Bonamia ostreae in the flat oyster, Ostrea edulis ArchiMer

Bonamia ostreae is an intracellular protozoan which is recognized as a cause of mortality in European populations of flat oysters (Ostrea edulis). Based on the recent characterization of actin genes of B. ostreae, specific primers were designed for real-time PCR using SYBR® Green chemistry. Specificity was demonstrated by the unique melting temperature peak observed in positive samples and by the lack of amplification in samples of oysters infected by closely related parasites, including Bonamia exitiosa. Molecular responses of Ostrea edulis haemocytes to an in vitro infection with Bonamia ostreae ArchiMer

Bonamiosis due to the parasite Bonamia ostreae is a disease affecting the flat oyster Ostrea edulis. B. ostreae is a protozoan, affiliated to the order of haplosporidia and to the cercozoan phylum. This parasite is mainly intracellular, infecting haemocytes, cells notably involved in oyster defence mechanisms. Suppression subtractive hybridisation (SSH) was carried out in order to identify oyster genes differentially expressed during an infection of haemocytes with B. ostreae. New evidence for the involvement of Paracartia grani (Copepoda, Calanoida) in the life cycle of Marteilia refringens (Paramyxea) ArchiMer

The dynamics of the protozoan parasite Marteilia refringens was studied in Thau lagoon, an important French shellfish site, for 1 year in three potential hosts: the Mediterranean mussel Mytilus galloprovincialis (Mytiliidae) the grooved carpet shell Ruditapes decussatus (Veneriidae) and the copepod Paracartia grani (Acartiidae). Parasite DNA was detected by PCR in R. decussatus. In situ hybridisation showed necrotic cells of M. refringens in the digestive epithelia of some R. decussatus suggesting the non-involvement of this species in the parasite life cycle. Ostreid herpesvirus 1 detection and relationship with Crassostrea gigas spat mortality in France between 1998 and 2006 ArchiMer
